ERG (ETS-related gene) AS A MARKER OF OPEN ENDOTHELIUM CAPILLARIES OF THE HUMAN SPLEEN

Abstract

Introduction. The views of researchers regarding the predominant type of microcirculation in the human spleen diverge significantly, even to the point of directly opposing statements. Thus, some scientists state that most of the blood passes through the open component of the microvasculature (Stainiger B. et all., 2021). While others consider that the closed type of blood circulation in the spleen reaches 80-90% of the blood flow (Buffet P. et all., 2011).

Purpose of the study. Definition of the ratio of open and closed components of blood microcirculation in the red pulp of the human spleen by using immunohistochemistry and morphometry methods     

Subjects and Methods. Immunohistochemical reaction with monoclonal antibodies CD31, CD34, Ki67, ERG was performed on paraffin blocks of the spleen from ten adults, from the fund of the Department of Anatomy of KazNMU named after. S. Asfendiyarova.  The volume density (Vv) of capillaries with open and closed ends on sections of the human spleen with IHC against ERG (ETS-related gene) was determined using an eyepiece grid at an objective magnification of 20 and 10. Calculations were carried out in 50 fields of view from each spleen. Statistical analysis was performedby using STATISTICA 7 (www.statsoft.com).

Result. Analysis of the preparations showed that only the ERG (ETS-related gene) marker most clearly and convincingly exhibits high expression in the endothelial cells of the microvascular bed of the normal human spleen.

In preparations with positive expression of ERG in endothelial cells, branches of terminal capillaries with open ends and blood cells extending into the surrounding red pulp were observed. Such capillaries could be traced over a long distance (up to 160 μm) right up to the open ends. As the open ends of the capillaries were approached, significant gaps were observed between adjacent endothelial cells.

Conclusion. The results of our study showed that the most highly specific marker for the morphological study of the microvasculature of the human spleen is normally the ERG (ETS-related gene) marker.

The main blood flow in the microcirculatory unit of the red pulp of the human spleen in performed research shows prevalence of a closed type, which is consistent with the data of Buffet P. et all. (2011) and do not confirm the opinion of Stainiger B. et all. (2021) supposing the predominance of open type of blood microcirculation in the human spleen.
